Civilization 5 - 1000+
Dragon Age Inquisition - 70 or so
Ultimate Marvel Vs Capcom 3 - 250+
SFIV - 250+
Quake 2, Battlefield 2, Tribes 1 + 2 easily in the multiple thousands when I was a student with no responsibilities or obligations back in the day.

MMOs, those fun, wretched time sinks. I've only stuck with two - WoW back when it released in 2005ish and more recently Eve which I stopped playing when the kid was on the way. Steam says I've notched 630 odd Eve hours. Eve is a beautiful, insanely deep game. If it didn't cost a monthly sub I'd keep checking in just to enjoy its ambience.
A recent SP game I've dropped the most time into would be Mass Effect 2 at 72 hours. Some console RPGs would be up there back in the day. I played Final Fantasy X inside out. I loved FEAR and have been through that game 4 or 5 times. HL2 is another multiple play-through. Halo CE is a big one too. Three play-throughs on XBox and another couple on PC.

Other than Destiny, I'd reckon the Mass Effect series has drawn the most time out of me. Each game has been finished multiple times and each original playthrough was about 50 hours.
Edit: Most time out of me in recent times... who knows what kind of hours were sunk into games when I wasn't an adult with responsibilities.

I couldn't count the hours of these games I played not only has a child but also as an adult...
Ladybug on Colecovision took away hundreds of hours of my life. I fucking love that game and I'm dying to play it again. Still have the Colecovision at my parents so I might few it up soon...

Resident Evil 4 (various incarnations). Sink most of my time on the import USA GameCube copy. My fav game of all time, I'd estimate close to, if not over a thousand hours on this as well! I've finished it so many times (on cube, wii, 360, ps3 and now PC). Never gotten bored of it.
Actually wish I had a proper timer for all the hours I sunk into my NES. Super Mario Bros took so much of my life from me. Even Aussie Rules Football on NES I played a tonne of. Good times.....
